Why this annotation
All top hub genes are MHC class II molecules (HLA-DRA, HLA-DPB1, HLA-DRB1, HLA-DRB5, HLA-DPA1, HLA-DQB1, HLA-DMA, HLA-DQA1) with strong enrichment in plasmablasts (2.6-6.7x). CD74 is the MHC-II chaperone invariant chain. LAPTM5, HCST, CD52, COTL1, LCP1 are immune activation/myeloid-associated genes enriched in plasmablasts. TRBC2 (T cell receptor beta) at 10.9x in plasmablasts likely reflects T cell contamination or doublets. CTSH is a lysosomal protease involved in antigen processing. This module represents a plasmablast state retaining MHC-II antigen presentation capacity, upregulated in inflammation. The plasmablast enrichment distinguishes this from mature plasma cells which downregulate MHC-II.
