Why this annotation
Hub genes are immediate early / NF-kB-driven activation genes: EGR4, EGR3, EGR2 (early growth response TFs), IER3, FOSL1, DUSP2 (AP-1/MAPK feedback), plus TNF, ICAM1, CD83, NFKBID, ZC3H12A (Regnase-1), RGS16 — all classic TCR/NF-kB/AP-1 activation program. This is a coherent early activation signature in CD4 T cells. The presence of EGR/IER/FOS family genes could indicate dissociation stress, but the strong NF-kB effectors (TNF, ICAM1, ZC3H12A, NFKBID, CD83) and disease-associated signal (inflammation_CD) support a genuine TCR/NF-kB activation program rather than pure artifact.
