ASCL2 is the master transcription factor for intestinal stem cell identity, directly driving LGR5+ ISC maintenance. RGMB modulates BMP signaling in the crypt niche. EPHB3 is a WNT target Eph receptor marking crypt progenitors. CDCA7 is a MYC target involved in cell cycle in stem cells. BEX2 and BEX4 are expressed in stem/progenitor contexts. PTPRO is a receptor tyrosine phosphatase expressed in ISCs. The module is strongly decreased in both UC and CD inflammation and strongly increased in remission — the canonical behavior of the ISC transcriptional program, which is suppressed during active IBD and restored upon remission.
