Why this annotation
LGR5 is the canonical marker of intestinal stem cells (Lgr5+ CBC stem cells). SMOC2 is a well-validated LGR5+ ISC co-marker. APCDD1 is a WNT target gene expressed in ISCs. PLEKHB1 and SLITRK6 are expressed in crypt base cells. PHLDA1 is a stress-responsive gene expressed in ISCs under injury. FN1 (fibronectin) may reflect niche matrix interactions. The module is strongly upregulated in inflammation and downregulated in remission — opposite to M71 (ASCL2). This paradox may reflect a stress-activated or injury-responsive LGR5+ ISC substate that expands during inflammation, distinct from the homeostatic ASCL2-driven ISC program. Very low mean expression and pct positive are consistent with a rare, highly specific ISC subpopulation.
