Why this annotation
Hub genes MX1, MX2, HERC5, PNPT1, SLFN5, TRIM25 are all well-established type I interferon-stimulated genes with direct antiviral functions. MX1 and MX2 are dynamin-like GTPases that restrict viral replication; HERC5 is an ISG E3 ligase for ISGylation; PNPT1 is involved in innate sensing of cytosolic RNA; SLFN5 restricts viral replication; TRIM25 is an E3 ligase that activates RIG-I antiviral signaling. TYMP (thymidine phosphorylase) has roles in nucleotide metabolism and is IFN-regulated. GPR180, POGLUT3, and PHF11 are less canonical but appear in IFN-stimulated transcriptomes. This module is a topological neighbor of M2, M66, and M127, all ISG modules, strongly confirming the classification.
