Why this annotation
The hub genes ISG15, IFI44, IFI44L, EPSTI1, SAMD9L, LY6E, PARP14, PARP9, XAF1, USP18, PLSCR1, DDX60, DTX3L are all well-established interferon-stimulated genes (ISGs) induced by type I and type III interferons. ISG15 is a ubiquitin-like modifier central to antiviral ISG responses; IFI44/IFI44L are classic ISG markers; SAMD9L, PARP9, PARP14, DTX3L, XAF1, USP18, DDX60, PLSCR1 are all canonical ISGs with antiviral functions. LGALS9 and NAMPT have immunomodulatory roles also linked to IFN signaling. HESX1 and HSH2D are less canonical but present in IFN-stimulated contexts. This module is a neighbor of M66, M127, and M164, all of which are also ISG/type I IFN modules, strongly supporting this classification. The uniform expression across subsets is consistent with a broadly active IFN response in pDCs, which are the primary producers of type I IFN.
