Why this annotation
The top hub HSPA5 (BiP/GRP78) is the master ER stress sensor and UPR initiator. DNAJB11 and DNAJC3 are ER-resident Hsp40 co-chaperones that activate IRE1/PERK branches of the UPR. ATP2A2 (SERCA2) maintains ER Ca2+ homeostasis and its disruption triggers ER stress. SPCS3 and TMED5 are ER membrane components. M6PR and BLOC1S6 link to lysosomal biogenesis downstream of UPR. The extreme mDC enrichment (60.7x) suggests this program is highly specific to monocyte-derived DCs, which are known to have high secretory/UPR activity. HSPA5's weak membership despite top PageRank indicates some heterogeneity, but the overall signature is consistent with UPR/ER stress. Neighboring M113 (ER chaperones) and M90 (ER translocon) support an ER-centric neighborhood.
