Why this annotation
All top hub genes are MHC class II molecules: HLA-DMB, HLA-DQB1, HLA-DPB1, HLA-DMA, HLA-DPA1, HLA-DRA, HLA-DRB1, HLA-DOA, HLA-DQA1 — a near-complete representation of the classical MHC-II locus. CD74 (MHC-II chaperone/invariant chain) and CIITA (master transcriptional regulator of MHC-II expression) are the canonical regulatory components. C2 (complement component 2, also MHC locus-encoded) rounds out the module. This is a pure, highly coherent MHC class II antigen presentation module driven by CIITA. Strong upregulation in IBD inflammation (UC: 1.820), reversal in remission. In the context of glia, this likely reflects IFN-gamma-induced expression of MHC-II on enteric glial cells, enabling antigen presentation. Neighbor to M73 (MHC-I) and M28 (IFN-gamma effectors) — together these modules form a complete IFN-gamma-driven antigen presentation axis across both MHC-I and MHC-II.
