Why this annotation
The top hub genes include JUNB, NR4A1, JUND, FOSB, FOS, ATF3, ZFP36, IER2, BTG2, DUSP2, PPP1R15A, and KDM6B — a classic immediate-early gene / AP-1 transcription factor response signature. NR4A1 and NR4A2 are nuclear receptors rapidly induced by stimulation. BTG2, ZFP36, DUSP2, and PPP1R15A are canonical early response genes. This pattern is consistent with cellular activation stress or dissociation-induced immediate-early gene induction (a well-known artifact in scRNA-seq), but in DCs it can also reflect genuine activation. The module is core coherence, uniformly expressed, and not enriched in inflammation. The neighbor context (M78 has heat shock genes) suggests a stress/activation neighborhood. The immediate-early gene signature in scRNA-seq of tissue DCs is most parsimoniously interpreted as dissociation stress artifact, though genuine DC activation cannot be excluded.
