Why this annotation

The top hub genes are HLA-A, HLA-C, HLA-B, and B2M — the core components of MHC class I antigen presentation machinery. IDO1 (indoleamine 2,3-dioxygenase, a key immunosuppressive/tolerogenic enzyme in DCs) and MREG (melanoregulin, involved in lysosomal/endosomal trafficking and cross-presentation) are also present. TXN (thioredoxin, redox regulation of antigen processing), RAB10 and RAB30 (vesicular trafficking), SPRED2 (negative regulator of MAPK), IL2RG (common gamma chain), ID2 (DC lineage transcription factor), EPSTI1 (interferon-stimulated), and TRADD (TNF signaling) round out the module. The uniform expression of HLA genes across subsets and the mDC enrichment of IDO1/MREG suggest this module captures MHC-I presentation combined with tolerogenic IDO1 activity in mature mDCs during inflammation. The inflammation upregulation and treatment decrease are consistent. Neighbor M68 covers maturation/migration; this module covers the antigen presentation arm of the same mDC activation state.
