Why this annotation
Hub genes CD80, CD274 (PD-L1), NECTIN2 (CD112), IL4I1, and KYNU are canonical markers of a tolerogenic/immunoregulatory mature DC state. CD80 and CD274 are co-stimulatory/co-inhibitory molecules upregulated on activated mDCs. NECTIN2 is a ligand for TIGIT/CD226 checkpoints. IL4I1 and KYNU are tryptophan-catabolizing enzymes that generate immunosuppressive metabolites (indole derivatives and kynurenine). DRAM1 links to autophagy-mediated antigen processing. TNFAIP8 is an anti-apoptotic NF-κB target. RIPK2, JAK2, LYN, and CSF2RB support inflammatory signaling. The strong enrichment in mDC, significant upregulation in inflammation (UC and CD), and the combination of checkpoint ligands with tolerogenic enzymes point to a regulatory/tolerogenic mDC activation state. Neighbor M101 is a strong IFN/GBP response module, consistent with a shared inflammatory mDC neighborhood.
