Why this annotation
Hub gene PADI4 is the master regulator of NET formation (citrullination of histones). ARG1 (arginase-1) is a canonical neutrophil maturation and immunosuppression marker. PLBD1 is a neutrophil granule protein. ITGAM (CD11b) mediates neutrophil adhesion and activation. APMAP and PPP2R5A are associated with myeloid differentiation. ADD3 and RIOK3 are expressed in mature neutrophils. The combination of PADI4 with ARG1, PLBD1, and ITGAM strongly points to a mature/primed neutrophil state poised for NET formation. Neighbor M3 reflects metabolic/housekeeping programs, and M36 reflects cytoskeletal programs — M6 is distinct in its NET-priming identity.
