Why this annotation
Top hub genes are STAT3 and STAT6, canonical JAK-STAT transcription factors mediating cytokine signaling (IL-6, IL-10, IL-4, IL-13). LRRFIP1 is a nucleic acid sensor that activates IRF3/beta-catenin innate immune pathways. KPNB1 (importin beta, nuclear import of STATs), EIF3A (translation initiation), GAK (clathrin-mediated endocytosis), ATP2A2 (SERCA2, ER calcium). Despite weak membership and uniform expression, the STAT3/STAT6 co-hub is biologically meaningful and points to a JAK-STAT cytokine response program, possibly at low basal activity across pDCs.
