Why this annotation
Hub genes are dominated by canonical immediate-early genes: FOS, FOSB, JUNB, JUN, JUND (AP-1 family), EGR1, ZFP36 (TTP, mRNA destabilizer co-induced with IEGs), IER2, and SOCS3 (cytokine signaling suppressor). HSPA1A and HSPA1B (HSP70) are stress-inducible chaperones co-regulated with IEGs under heat/proteotoxic stress. This is a textbook immediate-early gene / heat-shock response module. It is significantly down in inflammation and up in UC remission, consistent with loss of homeostatic IEG tone during active disease. The module is a topological neighbor of M72 (proteotoxic stress with chaperones) and M47 (sustained NF-κB/AP-1 inflammatory response), forming a coherent stress-to-inflammation axis in the network.
