Why this annotation
This module captures a second-wave or stronger antiviral ISG program, partially overlapping with M0 but enriched for IFIT family genes (IFIT1, IFIT3), RSAD2 (viperin), CMPK2, HERC5, MX2, HELZ2, and ISG20 — all strongly induced by type I IFN, particularly in robust antiviral responses. RSAD2/viperin and CMPK2 act together to deplete CTP for viral RNA synthesis. HERC5 is an ISGylation E3 ligase. ISG20 is an ISG with antiviral RNase activity. NT5C3A is involved in nucleotide metabolism and is IFN-regulated. PPM1K and HELZ2 are weaker ISGs. Compared to M0, this module may reflect a higher-magnitude or sustained interferon response.
