Why this annotation
This module is a textbook Type I Interferon response signature. All top hub genes are canonical ISGs (interferon-stimulated genes): RSAD2 (viperin), MX2, CMPK2, OAS3, IFIT1, USP18, PLSCR1, HERC6, HELZ2 — all directly induced by type I IFN signaling via the JAK-STAT1/2 pathway. LAMP3 is induced by IFN in dendritic cells and plasmacytoid DCs but also appears in activated T cells under IFN stimulation. LGALS9 (Galectin-9) is also upregulated by IFN and has immunoregulatory roles. The module is strong coherence with uniformly core/strong membership scores and very low mean expression (5.7% cells), consistent with a subset of MAIT cells undergoing antiviral/IFN activation. This is a highly confident, clean Type I Interferon module. Its neighborhood relationship with M58 (which contains STING1) is biologically coherent: STING activation upstream can drive type I IFN production, linking the two modules in an innate sensing → IFN response axis.
