Why this annotation
ATF6 is a canonical ER stress sensor and UPR transcription factor activated during proteotoxic stress. TMCC1 (transmembrane and coiled-coil domain 1) is an ER-shaping protein that maintains ER morphology. PACS1 is a multifunctional sorting protein that localizes to ER/TGN and regulates ER calcium homeostasis and the UPR. Together these three genes form a coherent ER stress/UPR core. MAP2K4 is a stress-activated kinase (JNK pathway) activated downstream of ER stress. GRB10 modulates IGF/insulin receptor signaling and is regulated by mTOR under stress. NCOA1 regulates transcriptional responses to cellular stress signals. DISC1 is a scaffold protein involved in intracellular signaling.
