Why this annotation
The top hub genes LDB3, DES, SPEG, and HSPB7 form a tight striated/cardiac muscle signature: LDB3 is a Z-disc scaffolding protein, DES is the canonical muscle intermediate filament desmin, SPEG is a striated muscle-enriched kinase essential for myofibril organization, and HSPB7 is a small heat shock protein expressed in cardiac and skeletal muscle. HACD1 functions in fatty acid elongation in muscle. KCNMA1 is a large-conductance calcium-activated potassium channel expressed in smooth muscle. The module is 'core' coherence with extremely low expression (0.008, 0.5% positive), uniform across fibroblast subsets, indicating contamination from muscularis propria smooth muscle or striated muscle cells rather than a fibroblast program. The increase in remission (delta_remission_UC = 0.157) may reflect restoration of muscularis integrity.
