Why this annotation
DDIT3 (CHOP) and DDIT4 (REDD1) are hallmark ISR/mTOR stress genes. SLC38A2 is an amino acid transporter that senses amino acid availability for mTORC1. MKNK2 phosphorylates eIF4E downstream of stress kinases. PMAIP1 (NOXA) is a pro-apoptotic BCL2 family member induced by ISR/CHOP. CEBPB is an inflammatory transcription factor also regulated by ISR. GLS (glutaminase) reflects metabolic reprogramming. IFRD1 is a stress-response coregulator. KLF4 is a transcription factor. MIDN activates the proteasome. This module represents the mTOR inhibition / amino acid deprivation arm of the integrated stress response, complementary to the ATF4-driven M46.
