Why this annotation
Hub genes FASLG (death ligand), SLAMF7 (NK activating receptor), and TIGIT (NK checkpoint receptor) define an NK cytotoxic activation program. ER chaperones PDIA3, CALR, P4HB, and PPIB support the secretory pathway required for cytotoxic effector production; CALR/PDIA3 also participate directly in NK cytotoxicity complexes. The module is significantly upregulated in active CD inflammation and downregulated with treatment, consistent with activated cytotoxic NK cells in intestinal inflammation.
