Why this annotation
This is the tightest and most interpretable module in the batch. Hub genes NFKB1 and NFKB2 are canonical NF-κB subunits. BCL3 is an IκB family member that modulates NF-κB transcriptional activity. OTULIN is a linear deubiquitinase that regulates NF-κB signaling by removing Met1-linked ubiquitin chains. PIM3 is a serine/threonine kinase promoting cell survival downstream of cytokine signaling. DUSP10 (MKP-5) is a MAPK phosphatase providing negative feedback. CSRNP1 is an NF-κB target stress-response transcription factor. BHLHE40 is induced by NF-κB and hypoxia. ZFP36 (TTP) destabilizes AU-rich mRNAs including TNF. NINJ1 mediates plasma membrane rupture in inflammatory cell death. The module is significantly upregulated in both UC and CD inflammation with treatment reversal, perfectly consistent with NF-κB-driven NK cell activation in IBD.
