Why this annotation

HOXA3, HOXA6, and HOXA9 are paralogous HOX cluster transcription factors that define definitive hematopoiesis and are essential markers of HSCs and multipotent progenitors (MPPs). HOXA9 in particular is a master regulator of HSC self-renewal and myeloid/lymphoid differentiation. AFF1 (AF-4) is a component of the super elongation complex and an MLL fusion partner in ALL, expressed in lymphoid progenitors. ARHGAP25 is a Rho-GAP expressed in myeloid progenitors regulating phagocytosis. ST3GAL1 mediates sialylation critical for HSC niche retention. LPIN1 regulates lipid metabolism in progenitors. ADGRE2 is a myeloid-expressed adhesion receptor. JAML mediates leukocyte adhesion. PRKCB is expressed in B-cell progenitors. The HOX cluster signature is the anchor of this module, indicating a definitive HSC/MPP program.
