Why this annotation
GUCY1A1 (soluble guanylate cyclase alpha-1) is highly expressed in megakaryocytes and platelets, where cGMP signaling regulates platelet function. IQGAP2 is a scaffold protein enriched in megakaryocytes and platelets, regulating cytoskeletal organization during platelet biogenesis. These two genes anchor the module to the megakaryocyte lineage. However, GBP4, IFI16, TRIM22, and HLA-E are classical interferon-stimulated genes (ISGs), suggesting an interferon-primed substate co-expressed in this population. ELK3 (NET/SAP-1) is an ETS transcription factor active in megakaryopoiesis. MAML2 is a Notch co-activator. The module likely represents an interferon-stimulated megakaryocyte-biased progenitor or MkP (megakaryocyte progenitor) state. The neighbor context (surrounded by HSC/progenitor modules) supports this being a lineage-committed HPC substate rather than contamination.
