Why this annotation
Hub genes include multiple HOXA cluster members (HOXA3, HOXA5, HOXA7, HOXA9) alongside metabolic enzymes of the TCA cycle (SUCLG2, NDUFV1, CYCS), detoxification (CAT, PTGR1, ADH6), and PEBP1/DDC as colonocyte differentiation markers. The strong negative delta_inflammation in both UC and CD indicates this program is lost during intestinal inflammation, consistent with a normal colonocyte identity/differentiation state. HOX genes define posterior gut positional identity and are known to be expressed in mature colonocytes. The metabolic genes (PCCA, BDH2, SUCLG2) reflect the oxidative metabolism of differentiated colonocytes. Neighbor M130 also contains HOX genes (HOXB13, HOXD11, HOXD13), reinforcing a shared colonocyte positional identity theme. M65 emphasizes the metabolic/mitochondrial aspect of this differentiated state.
