Why this annotation
IRF7 and IRF9 are canonical type I interferon pathway transcription factors. MAP3K8 (COT1/TPL2) is an inflammatory kinase downstream of TLR/TNF signaling. IL12RB1 encodes the IL-12 receptor beta-1 chain, a hallmark of ILC1/NK cells and their inflammatory activation. SLC2A3 (GLUT3) upregulation supports metabolic reprogramming during immune activation. Several genes (GRAMD2B, SH3TC1, APMAP, TSEN54, SLC2A3, OTULINL) show ILC1 enrichment. IRF9 is the ISGF3 component linking IFN-γ and type I IFN responses. Despite weak coherence, the IRF7/IRF9/MAP3K8/IL12RB1 axis points to an IFN-driven inflammatory activation program enriched in ILC1.
