Why this annotation
Hub genes NR4A1, JUN, ATF3, KLF6, GADD45B, IER5, ZFP36L1, PPP1R15A, SERTAD1, RGCC are immediate early gene/stress-response transcription factors. NR4A1, JUN, ATF3, KLF6 are classic immediate early genes induced rapidly by TCR stimulation, oxidative stress, or dissociation. PPP1R15A (GADD34) is a stress-response phosphatase. ZFP36L1 is an mRNA decay factor induced by activation. GADD45B is a DNA damage/stress response gene. IER5 is an immediate early response gene. This pattern is consistent with either TCR activation-induced early response or dissociation stress. As a topological neighbor of M62 (also immediate early/AP-1), both modules together strongly suggest this is the immediate early activation/stress response program. The presence of EIF4A2 and EIF5 (translational regulators) and ARRDC3 (stress-responsive arrestin) further supports a stress/activation response rather than pure dissociation artifact, though dissociation stress cannot be excluded.
