Why this annotation
PROX1 is the master transcription factor for lymphatic endothelial identity and is essential for lymphatic specification. ARHGAP29 is a Rho GTPase-activating protein expressed in lymphatic endothelium and regulates lymphatic valve morphogenesis. CALD1 (caldesmon) is a cytoskeletal regulator expressed in lymphatics. NFIC is a transcription factor involved in lymphatic development. VPS36 (ESCRT-II), RAB11FIP1 (endosomal recycling), and STX7 (vesicle trafficking) suggest active vesicular transport. RFC1 and BOD1L1 relate to chromatin/DNA maintenance. The PROX1-anchored signature with ARHGAP29 strongly points to lymphatic identity/specification. Not downregulated in inflammation, consistent with constitutive identity program.
