Why this annotation
Hub genes are canonical immediate early genes (FOSB, FOS, JUNB, JUN, JUND, EGR1) co-expressed with flow-responsive KLFs (KLF2, KLF4), heat shock proteins (HSPA1A/B, DNAJB1, HSP90AA1), and negative regulators of inflammation (ZFP36, DUSP1, SOCS3). NR4A1 and KLF2 are well-established shear stress sensors in endothelial cells. The module is significantly suppressed during UC inflammation and restored in remission, consistent with loss of laminar flow during mucosal inflammation and recovery of homeostatic mechanosensory signaling. HES1 and MIDN are also IEG/stress-response genes. All 20 genes fit a shear stress-induced immediate early gene program.
