Why this annotation
Hub gene LAT (linker for activation of T cells) is the central scaffold of the TCR signaling complex. Supporting genes include RASGRP2 (RAS guanyl nucleotide-releasing protein 2, downstream of TCR), PLEK (pleckstrin, DAG effector in TCR signaling), MATK (megakaryocyte-associated tyrosine kinase, CSK-like inhibitor of Src kinases), FGR (Src-family kinase), BIN2 (bridging integrator), TNFRSF14 (HVEM, costimulatory/coinhibitory), TSPAN32, ADGRG5, and GPSM3. The co-expression of LAT with RASGRP2, PLEK, and MATK strongly supports a TCR proximal signaling program. The module is tightly coherent (core) and uniformly expressed across subsets, consistent with a basal TCR signaling state.
