Why this annotation

The top hub genes are CXCL9, CXCL10, CXCL11 (IFN-gamma-induced chemokines), GBP1/2/4/5 (guanylate-binding proteins, canonical IFN-gamma effectors), TAP1 (antigen presentation, IFN-gamma target), SAMHD1, APOL6, TRIM22, PARP14, EPSTI1, IL18BP, IDO1, CASP1, and UBD — all well-established IFN-gamma-stimulated genes. This is a tight IFN-gamma response signature, distinct from the type I IFN module (M1) which is its topological neighbor. The strong upregulation in UC and CD inflammation (delta_inflammation_UC=0.875, delta_inflammation_CD=1.348) and reversal in UC remission (-1.247) is fully consistent with IFN-gamma-driven endothelial activation in IBD. The neighbor module M1 captures type I IFN (IFIT/MX/OAS genes), while M87 captures the IFN-gamma arm — both are interferon response programs but driven by distinct cytokines, explaining their co-expression network proximity. Concordant genes: CXCL9, CXCL11, GBP1, GBP5, CXCL10, SAMHD1, TAP1, APOL6, SECTM1, TRIM22, PARP14, IL18BP, GBP2, EPSTI1, GBP4, ISG20, UBD, CASP1, IDO1 (19 of 20 listed; APOL2 is weakly associated but APOL family members are IFN-gamma inducible).
