Why this annotation

The module is dominated by chromatin-modifying enzymes: KMT2C and KMT2E (H3K4 methyltransferases), ASH1L (H3K36 methyltransferase), SETD5 (H3K36 methyltransferase), JMJD1C (H3K9 demethylase), KDM2A (H3K36/H3K4 demethylase), KAT6A (H3K9/K14 acetyltransferase), EPC1 (NuA4/Polycomb-associated), EHMT1-neighbor context. TET2 is a canonical myeloid DNA demethylase critical for macrophage epigenetic programming. AFF4 (super elongation complex), AGO3 (RNA silencing/Argonaute), RC3H1 (Roquin mRNA stability regulator), and RBM47/RBM33 (RNA binding) add post-transcriptional regulation. This is the clearest module in the batch: a chromatin and epigenetic remodeling program highly characteristic of monocyte-derived macrophages, where TET2 and KMT2 family members are well-established regulators of myeloid identity and inflammatory gene expression.
